Py-Dyes are a class of amine-reactive dyes that possess unique physical and spectral properties that make them ideal for use in general-purpose protein labeling. Naturally amine reactive, the unbound form of a Py-Dye has a quantum yield of nearly zero. However, upon conjugation the dye short wave shifts and acquires a high quantum yield. The result is a family of dyes that do not require complicated conjugation chemistry, and that provide high-intensity signals with almost no background interference. Thus, Py-Dye members are used in both the ProStain™ Protein Quantification Kit and the CE Dye Kits for capillary electrophoresis. To find out more about Py-Dyes, click here.

In addition to intensity-based measurements, fluorescent signals can be measured using parameters such as lifetime or polarization. These methods overcome problems associated with auto-fluorescence and background fluorescence because these signals decay rapidly, and because polarized signals are highly specific to the fluorophore being measured. Sensitivity is enhanced because fluorescent signals are read over longer, time-gated periods that amplify signal intensity. Active Motif Chromeon offers a line of RuLabels that are ready for conjugation to biomolecules. These dyes emit polarized light with a long decay time between 400 ns and 5 µs, making them ideal for use in a wide variety of methods. To find out more, click here.
